In vitro
A66 is a high selective PI3K inhibitor which can inhibit PI3K-alpha with 32 nM. A66 inhibits p110-alpha E545K and p110-alpha H1047R with IC50 of 30 nM and 43 nM. A66 shows strong inhibition to p110-alpha and little inhibition to other PI3K isoform. A66 can inhibit cell lines harboring H1047R mutations in PIK3CA and have high levels of p110-alpha and class-Ia PI3K activity.
